Kind of an odd question - most American.
Smokeless powder was a French development, and black powder finds its roots in Asia, so we can argue every cartridge has some foreign roots if we go back far enough.
Almost all cartridges are developed and derived from concepts borrowed from previous generations of cartridges, foreign and domestic.
For example,
The Boxer primer we currently use in our shells is a European concept.
The Berdan priming they use, is an American concept.
For me, the most American cartridge I have killed a deer with is:
45-70
It was developed by the US military.
It was conceived and created prior to the advent of either priming system mentioned above.
It is early enough that it did not borrow a lot from other countries developments, however the features of the shell are not unique to American cartridges of the era.
I guess I need to take something with a Burnside or Maynard cartridge.
Is that a good enough reason to add another piece to the collection?

If it wasn't specific about cartridge it would be the old sidelock octagon barrel .45 caliber with patched roundball sitting on FFG black powder handed down through the family on my fathers side.
45-90 Winchester out of an 1886 Winchester rifle made in 1893, with an original Winchester factory round... but I don't know when it was loaded, turn of the century I would think. Gotta love a big old charge of black!!!
Birds, 12 and 20 gauge. Small game .22 short hp. Everything else has been, from start to now, 30-30, .308, 30-06, .270 and 6.5 Creedmoor. From 1984 through 2020 was strictly the .270. Don’t know which would be purely American but I am and I own them, so there is that.
