EDIT: Nope, must be unloaded unless you have a permit. Even if you have a permit, it can only be magazine loaded, not chamber loaded (see section 1289.13 of the sda lawbook).

If you have a CHL, you can carry a rifle or shotgun magazine loaded in your vehicle. Empty chamber full magazine.
If you don't have a carry license you can still transport a rifle, pistol, or shotgun, it just has to be unloaded (i.e, no ammo in the magazine or chamber).
Any person, except a convicted felon, may transport in a motor vehicle a rifle, shotgun or pistol, open and unloaded, at any time. For purposes of this section "open" means the firearm is transported in plain view, in a case designed for carrying firearms, which case is wholly or partially visible, in a gun rack mounted in the vehicle, in an exterior locked compartment or a trunk of a vehicle.

Any person, except a convicted felon, may transport in a motor vehicle a rifle or shotgun concealed behind a seat of the vehicle or within the interior of the vehicle provided the rifle or shotgun is not clip, magazine or chamber loaded. The authority to transport a clip or magazine loaded rifle or shotgun shall be pursuant to Section 1289.13 of this title.

Any person who is the operator of a vehicle or is a passenger in any vehicle wherein another person who is licensed pursuant to the Oklahoma Self-Defense Act to carry a handgun, concealed or unconcealed, and is carrying a handgun or has the handgun in such vehicle, shall not be deemed in violation of the provisions of this section provided the licensee is in or near the vehicle.

That's actually one place where the SDA took a step back--before it was enacted, you could legally transport a magazine-loaded rifle or shotgun...

The one thing to bear in mind is if you permanently carry a magazine loaded firearm concealed in your vehicle, make sure no one without a permit is allowed to operate your vehicle.

If a spouse or child is driving your vehicle and doesn't have a permit, it won't matter if they didn't know about the firearm mounted under the back seat or not, they can be charged for this.

For example if your child is pulled over by a cop and they ask them, You don't mind if i check your car do you?

This is good advice. It's also a good reason why your kids should be educated on the 4th and 5th amendments before they are ever left home alone, or are old enough to drive. "Officer, I don't consent to any searches, and I don't want to answer any questions. Am I free to go?" Rinse and repeat.
