I've noticed on some of the NightForce optics, they have "Hi-Speed turrets" whereas others do not. Is this a major significance, other than a difference of $200+ on the price?

I'm asking because I've been looking at scopes and I'm unsure of which to purchase, either with the Hi-Speed or without.

All Nightforce scopes come with target turrets. In other words, they're clearly marked and you can easily adjust them by hand rather than having to get a coin out to adjust them. On some, there is no cover on the turrets and I presume this is what is called high speed but I could be wrong. On others, specifically the benchrest models, there is a cover that screws on over the turrets that has to be removed before you can make an adjustment.

Personally, I would go for high speed turrets. You could also call Nightforce or your potential Nightforce dealer for a more official answer to your question.
